石蠟制片法是將材料經(jīng)固定、脫水、透明、浸蠟后包埋在石蠟里面進(jìn)行切片的方法。此法適用范圍，制片手段完備，能將材料切成極薄的片子（可切至2-8微米左右），并能制成連續(xù)的片子，這是其它制片法難以達(dá)到的，因此在光學(xué)顯微鏡的制片技術(shù)上它是zui常用的一種方法。除了一些經(jīng)不住石蠟切片法中所應(yīng)用的各種藥劑處理的材料不能應(yīng)用石蠟切片以外，一般的材料都可以用此法制片，但有制作時(shí)間較長(zhǎng)，操作過(guò)程復(fù)雜以及材料易發(fā)硬或發(fā)脆等缺點(diǎn)。該法多采用旋轉(zhuǎn)式切片機(jī)進(jìn)行切片。
石蠟制片操作程序如下：
（一）材料的采集與分割
采集材料應(yīng)根據(jù)制片的目的和要求而定，材料要有代表性，無(wú)病蟲(chóng)害或其它損傷，如要觀察病理解剖，則要取病斑、病癥部位。采下的材料應(yīng)立即放在標(biāo)本箱或濕布包起來(lái)帶回實(shí)驗(yàn)室進(jìn)行分割固定（如有條件也可在試驗(yàn)地采集后進(jìn)行分割固定）。為使固定液能迅速的滲透材料，材料要進(jìn)行分割。分割時(shí)動(dòng)作要迅速，以防材料萎蔫。分割塊的大小，宜小不宜大，一般不超過(guò)1cm3，分割后立即殺死固定。
